Research progress on the relationship between ceramide and diabetes mellitus and its complications

Abstract:Globally, 1 in 11 adults has diabetes, and the threat of diabetes to the human body is mainly its serious, sometimes lifethreatening complications. Ceramide is a sphingolipid substance that plays a key role in the regulation of signal transduction pathways, which is mainly synthesized by three pathways: de novo synthesis pathway, sphingomyelin hydrolysis/degradation pathway and recovery/remediation pathway. Ceramide has been linked to the development of diabetes by inducing β cell apoptosis, insulin resistance, and inhibiting insulin gene expression. Some species or ceramide proportions of ceramide may serve as biomarkers of diabetes risk, and understanding the role of ceramides in diabetes and its complications is critical for treatment planning.This review discusses the relationship and mechanism of ceramides in the progression of diabetes and its complications, with a focus on the mechanisms of correlation between ceramides and diabetic retinopathy, diabetic nephropathy, diabetic cardiovascular disease, and diabetic neuropathy, as well as some potential and emerging treatment strategies.
